The b-galactoside-binding activity of oxidized rhGAL-1 (10 µg) was determined by lactose–agarose affinity chromatography under nonreducing (A) or reducing (B) conditions. Protein elution with a linear gradient of lactose was monitored by measuring A215. Oxidized rhGAL-1 did not bind to a lactose–agarose column under nonreducing condition.
